What is Doxazosin mesylate  ?

Doxazosin mesylate is mainly used in patients with primary mild to moderate hypertension and hypertension with benign prostatic hypertrophy. Doxazide can be used in combination with thiazide diuretics and β -blockers, calcium antagonists, or angiotensin converting enzyme inhibitors for patients whose blood pressure is difficult to control with a single drug.

Doxazosin mesylate pharmacology toxicology :

This product is a selective postsynaptic α 1-adrenal receptor blocker. By blocking α1- receptor to achieve vascular dilation, reduce vascular resistance, reduce the role of blood pressure. The antihypertensive effect was maintained for 24 hours, and the maximum antihypertensive effect appeared 2 ~ 6 hours after taking the medicine. Unlike non-selective α -blockers, no resistance was observed with long-term use, tachycardia was rarely observed during maintenance therapy, and plasma renin was elevated.

By selectively blocking α -adrenergic receptors in the prostate smooth muscle matrix, capsule, and bladder neck, the product improves urodynamics and clinical symptoms in symptomatic patients with prostatic hyperplasia.

Pharmacokinetics of doxazosin mesylate :

After oral administration, the product is absorbed rapidly, the peak time is 2-3 hours, the bioavailability is about 65%, the protein binding rate is 98%, and the terminal elimination half-life (T1/2) is 19-22 hours. The absorption delay was about 1 hour after eating, but the clinical effect was not significantly reduced. This product is widely metabolized in the liver. Although several active and inactive metabolites have been identified, their pharmacokinetic properties remain unclear. This product is mainly excreted by feces, 63% are metabolites and 4.8% are prototypes; Renal excretion 9%. The area under the curve of morning administration was 11% lower than that of evening administration, and the peak time was significantly lower than about 2 hours in the evening. Steady-state blood drug concentration studies in hypertensive patients showed a linear relationship between doxazosin dosage and efficacy, and doxazosin dosage was 2-16mg per day. Pharmacokinetic studies showed that the plasma half-life and clearance of doxazosin were not affected by age or renal impairment, but increased by 40% in cirrhotic patients with a single oral dose of doxazosin 2mg. At present, there are few data about the effect of doxazosin on liver metabolism.

In uncontrolled clinical trials of hypertension, the most common adverse reactions were dizziness, headache, fatigue, weakness, postural dizziness, vertigo, edema, drowsiness, nausea, and rhinitis. Postural syncope is rare, and urinary incontinence is rarely reported.

Allergic to quinazoline (such as prazosin, terazosin) and severe hypotension after taking this product should not be tolerated.
