Ponazuril is an antiprotozoal drug used in animals to control infections caused by protozoa. It is also considered a coccidiostat. Ponazuril (also known as toltrazuril sulfone) is a metabolite of the poultry antiprotozoal drug toltrazuril. Ponazuril is a triazine-based drug that acts to inhibit enzyme systems in protozoa or decreasing pyrimidine synthesis. It is specific for apicomplexan organisms because the action attacks the apicoplast organelle in protozoa. This action produces a specific effect as an antiprotozoal agent without affecting other organisms or the treated animal. It is approved for use in horses but also has been used in puppies, kittens, exotic, and zoo animals for treatment of coccidial infections.

Pharmacokinetics: Ponazuril has high, but variable, oral absorption in horses. The half-life data for horses has been conflicting with different studies reporting 1.6 days, 2.5 days, and 4.5 days. Ponazuril produces concentrations in cerebrospinal fluid (CSF) of horses that are 3.5% to 4% of serum concentrations but high enough to inhibit protozoa. One week of daily dosing in horses is needed to reach steady-state concentrations. A level of 100 ng/mL is necessary to prevent and treat infections caused by Sarcocystis neurona. The half-lives are 2.4 hours in cattle and 5.6 days in pigs.

Ponazuril is used for protozoa such as Isospora, Coccidia spp., Toxoplasma gondii, Sarcocystis neurona, and Eimeria spp. It is useful in treating Neospororsi or Toxoplasmosis, EPM, Coccidiosis, and Cryptosporidium spp. Ponazuril requires a prescription from your veterinarian.

Are There Any Risk Factors Associated With This Medication?

If any of the following serious side effects occur, stop giving Ponazuril and seek emergency veterinary medical attention; an allergic reaction (difficulty breathing; swelling of the lips, tongue, or face; hives), scratching, sudden onset of diarrhea, vomiting, shock, seizures, pale gums, coma.

Discontinue giving Ponazuril and contact your veterinarian if your pet develops; poor appetite, drowsiness, diarrhea, labored breathing, weakness, incoordination, fainting, accumulation of fluid in the lungs or abdomen, and cough. Other side effects may also occur.

Talk to your veterinarian about any side effect that seems unusual or bothersome to the pet.

Storage

Ponazuril capsules, powder, and paste should be stored at room temperature in a tightly sealed container and kept away from direct sunlight.

Liquid Ponazuril should also be stored at room temperature. Shake well before administering the dosage.

Medication should not be ingested by humans. In homes with children, a childproof container is recommended. If a human accidentally swallows this medication, call your doctor right away.
